SYNTHESIS OF ZnO NANOPARTICLES AND THE ADVANTAGEOUS RESISTANCE ON THE MULTIDRUG RESISTANCE IN ACINETOBACTER BAUMANNII AND STAPHYLOCOCCUS AUREUS ISOLATES

ABSTRACT : ZnO NPs were synthesized by direct precipitation method using Zinc nitrate and Potassium hydroxide KOH. Xray diffraction (XRD), Transition electron microscopy (TEM), Atomic Force Microscopy (AFM) and UV- visible spectrophotometer were used to inspect and study ZnO nanoparticles properties. X-ray analysis has verified the formation Zinc Oxide nanoparticles. The average crystallite sizes have been determined to be 30.6 (nm). TEM measurement confirmed ZnO nanoparticles are close to hexagonal in shape. AFM confirmed the average size of the Zinc Oxide nanoparticles was 57 (nm) and the average surface roughness obtained was 5.59 (nm). UV-Vis spectrum exhibits the absorption peak of Zinc Oxide nanoparticles at 375 nm. In our study, rate of occurrence of Acinetobacter baumannii and Staphylococcus aureus isolated from different clinical specimens have been investigated, also, sensitivity of the multi drug resistance Acinetobacter baumannii and Staphylococcus aureus to ZNO-NPs. In current study, bacteria obtained as follows: (21.35%) of Staphylococcus aureus and (13.52%) of A. baumannii were isolated from blood and (6.72%) of S. aureus and (5.22%) of A. bumannii were isolated from urine. It founds (46.15%) isolates of Acinetobacter baumannii and 38.46% S. aureus isolates were multi drug resistance (MDR). These isolates showed the strong resistance against most of antibiotics used. This study found that the bacterial isolates are highly sensitive to high concentrations of ZnO-NP this gives large zone of inhibition. Different concentrations of ZnO-NPs mixed with ceftriaxone (2.5mg/ml, 5mg/ml) were studied by using well diffusion method.
